Animal Models Spontaneously Regressing Malignant Melanomas

چکیده

Human melanoma is the fastest growing cancer currently in USA and worldwide. Melanoma accounts for 5% of all skin cancers but 71% of all skin cancer deaths. The risk for melanoma has steadly increased since 1935 when the risk was 1:1,500. Today one in 74 people are diagnosed with melanoma. The mechanism for melanoma initiation is unknown and no significant advances in medical therapies (survival) for patients with advanced melanoma have occurred in the past 30 years. Additional research and drug development are required. Miniswine offer insights into the genetics, immunology and pathogenesis of malignant melanoma.

Development of a new melanoma model in C57BL/6 mice.

In the present study, we induced melanomas in C57BL/6 mice by a single application of 7,12-dimethylbenz(a)anthracene to the scapular region of 4-day-old mice, followed by twice-weekly applications of croton oil. Of 20 mice treated, melanomas arose in two female littermates. The first melanoma (JB/MS) arose 16 weeks after initiation of treatment, and the second melanoma (JB/RH) arose 23 weeks la...

Murine metastatic tumour models for cancer gene therapy research.

The appropriate use of animal tumour models has a pivotal role in the evaluation of any new anti-cancer therapy. Indeed, animal models of human diseases have been emphasised in the early development and evaluation of gene therapy. Given that most cancer treatment failures and cancer-related mortality are the direct results of metastatic cancers, the increasing use of clinically-relevant metasta...

Comparative Aspects of Canine Melanoma

Melanomas are malignant neoplasms originating from melanocytes. They occur in most animal species, but the dog is considered the best animal model for the disease. Melanomas in dogs are most frequently found in the buccal cavity, but the skin, eyes, and digits are other common locations for these neoplasms. The aim of this review is to report etiological, epidemiological, pathological, and mole...

Ladarixin, a dual CXCR1/2 inhibitor, attenuates experimental melanomas harboring different molecular defects by affecting malignant cells and tumor microenvironment

CXCR1 and CXCR2 chemokine receptors and their ligands (CXCL1/2/3/7/8) play an important role in tumor progression. Tested to date CXCR1/2 antagonists and chemokine-targeted antibodies were reported to affect malignant cells in vitro and in animal models. Yet, redundancy of chemotactic signals and toxicity hinder further clinical development of these approaches. In this pre-clinical study we inv...
